Neurologische Erkrankungen können oft nur unzureichend pharmakologisch behandelt werden, da entweder wirksame Medikamente fehlen oder vorhandene Arzneimittel starke Nebenwirkungen hervorrufen. Mit den klassischen Methoden der Arzneimittel-Entwicklung wird in der Regel nach niedermolekularen Wirkstoffen gesucht, welche die in der Erkrankung involvierten Systeme aktivieren oder inhibieren. Leider war dieser Weg bisher häufig erfolglos, da die in neurologischen Erkrankungen betroffenen Systeme sehr oft elementare Hirnfunktionen regulieren. Deren globale Aktivierung oder Inaktivierung ist daher mit starken Nebenwirkungen behaftet. Idealerweise sollten Arzneimittel nur in erkrankten Nervenzellen wirken, während gesunde Nervenzellen möglichst unbeeinflusst bleiben. In diesem Forschungsprojekt wird ein neuer Ansatz evaluiert, mit dem es möglich wäre sich diesem Ideal anzunähern.

Das Zielprotein dieses Projekts ist ein Rezeptor für den inhibierenden Neurotransmitter GABA: der GABAB-Rezeptor. GABAB-Rezeptoren kommen praktisch in allen Neuronen vor, regulieren alle wesentlichen Hirnfunktionen und zeigen in neurologischen Erkrankungen häufig eine verminderte Funktion. Dies führt zu einer Störung der Balance zwischen Erregung und Inhibition in neuronalen Netzwerken und ist eine wichtige Komponente neurologischer Erkrankungen. Daher wäre eine pharmakologische Beeinflussung dieser Rezeptoren als therapeutischer Ansatz vielversprechend. Leider war dieser Weg mit der klassischen Methode bisher weitgehend erfolglos. In diesem Projekt verfolgen wir eine alternative Strategie, bei der wir zunächst die Mechanismen untersuchen, welche die GABAB-Rezeptoren unter pathologischen Bedingungen regulieren. Da die Kommunikation der Rezeptoren mit ihrer zellulären Umgebung weitgehend durch Interaktionen mit anderen Proteinen erfolgt (Protein-Protein-Interaktionen), versuchen wir diejenigen Interaktionen aufzuspüren, die für die pathologische Regulation der Rezeptoren verantwortlich sind. Bisher haben wir vier mögliche Kandidaten solcher Mechanismen identifiziert. In der ersten Phase des Projekts werden wir anhand von neuronalen Zellkulturen erkrankungsrelevante Protein-Interaktionen des GABAB-Rezeptors im Detail analysieren. Diese Informationen werden es uns ermöglichen kurze synthetische Peptide zu entwickeln, welche die Protein-Interaktionen mit dem Rezeptor verhindern, so dass eine pathologische Regulierung des GABAB-Rezeptors unterbunden wird. Diese Vorgehensweise hat den Vorteil, dass es nicht zu einer globalen Aktivierung der GABAB-Rezeptoren kommt, sondern lediglich der normale Zustand in den erkrankten Nervenzellen wiederhergestellt wird. In der zweiten Phase des Projekts soll die Wirksamkeit dieser Peptide anhand zweier Mausmodelle für neurologische Erkrankungen getestet werden. In einem Mausmodel für den Schlaganfall erwarten wir eine neuroprotektive Wirkung der Peptide, d. h. eine Verhinderung des Absterbens von Nervenzellen. In einem Mausmodel für Drogensucht erwarten wir eine Reduktion des Drogenkonsums sowie eine Verminderung der Entzugsymtome. Wenn sich unsere Hypothesen bestätigen, bilden die Ergebnisse dieses Projekts die Grundlage für die Entwicklung hochspezifischer therapeutischer Ansätze zur Bekämpfung neurologischer Erkrankungen.

Although most biological processes are primarily regulated via protein-protein interactions, drug discovery largely relies on the development of small molecules that target drug binding sites present on a relatively small number of proteins. In the field of neurological disorders, targeting the binding sites of major neurotransmitter receptors is often not a feasible strategy because they are involved in many basic brain functions. Consequently, activating or inhibiting these receptors inevitably will induce a plethora of side effects. A promising solution to this problem might be the targeting of disease-specific protein-protein interactions.One neurotransmitter receptor that is ubiquitously expressed in the central nervous system and is involved in all major brain functions and many neurological diseases is the G protein-coupled GABA(B) receptor, which control neuronal excitability by mediating inhibition. There is accumulating evidence that cell surface expression of the receptors and thereby GABA(B) receptor-mediated inhibition is compromised under pathological conditions, contributing to the disease state. Therefore, we aim at identifying specific mechanisms affecting cell surface expression of the receptors under pathological conditions. This knowledge will enable us to design small interfering peptides targeting protein-protein interactions that cause the aberrant receptor regulation to restore normal receptor expression and thereby GABA(B) receptor-mediated inhibitory control. We expect that this strategy specifically target those GABA(B) receptors involved in the disease state and leaves receptors in healthy neurons largely unaffected. However, we have currently only limited knowledge on the mechanisms regulating cell surface expression of GABA(B) receptors. Therefore, we plan in the first part of the proposal to continue our efforts delineating the mechanisms that determine cell surface expression of GABA(B) receptors using cultured neurons. Our current findings suggest that the availability of functional GABA(B) receptors is regulated by multiple pathways in different cellular compartments in response to the level of neuronal activity. We intend to focus on the analysis of four distinct mechanisms:Subproject 1: The number of receptors in the ER available for forward trafficking to the cell surface is regulated by proteasomal degradation and most likely by interaction with the ER resident protein PRAF2. We intend to identify the pathway(s) linking neuronal activity to proteasomal degradation of GABA(B) receptors.Subproject 2: Protein phosphatase 2A (PP2A) mediates downregulation of cell surface GABA(B) receptors under pathological conditions but the mechanism involved is unknown. Here we intend to identify the underlying mechanism, which is most likely receptor endocytosis or recycling.Subproject 3: Lysosomal degradation of internalized receptors is regulated by phosphorylation-induced ubiquitination of GABA(B1) involving CaMKIIb and probably ERK II. Here we plan to analyze the role of ERK II in this mechanism.Subproject 4: There is evidence that the heterodimerization of cell surface GABA(B) receptors can be affected by a so far unknown mechanism. Here we intend to test our hypothesis that monoubiquitination via the E3 ligase MARCH1 controls heterodimerization of cell surface GABA(B) receptors.Using the information gained in these subprojects we will develop cell-penetrating peptides that interfere with critical protein-protein interactions in the distinct mechanisms to prevent downregulation of GABA(B) receptors under pathological conditions. The in vitro validated and optimized interfering peptides will then be used in the second part of the proposal to analyze the contribution of these mechanisms to neurological disorders. We intend to employ mouse models of two distinct diseases: 1. cerebral ischemia as an example of severely impaired excitation/inhibition balance associated with profound downregulation of total GABA(B) receptor expression and 2. methamphetamine-induced addiction as an example of drug-induced neuronal plasticity that appears to affect selectively cell surface levels of GABA(B) receptors with no obvious effect on total receptors. We will analyze whether restoring normal GABA(B) receptor expression by interfering with the relevant protein-protein interactions is a valid strategy for the development of novel, highly specific therapeutic approaches.
