Die biologischen Zellen sind von einer hauchdünnen Membran umgeben. Diese Zellmembran besteht aus 2 aufeinander liegenden Phospholipischichten. Welche Kräfte genau zwischen diesen zwei Schichten aktiv sind, ist noch weitgehend unbekannt. Die vorliegende Arbeit wird grundlegend untersuchen, was geschieht, wenn die beiden Phospholipidschichten miteinander verbunden werden und so ihrer Unabhängigkeit beraubt werden.

Inhalt und Ziel des Forschungsprojekts 
Das Ziel dieser Arbeit ist es, verschiedene neue Klassen von künstlichen Phospholipiden zu synthetisieren. Diese Phospholipide werden dann in biophysikalischen Untersuchungen von Lipid-Monoschichten und Bischichten verwendet werden. Die Moleküle sind dergestalt, dass über verschieden chemische Interaktionen zwei Lipidschichten miteinander verbunden werden können, entweder über Wasserstoffbückenbindungen oder über eine Polymerisation.
Durch die Verbindung der beiden Lipidschichten einer Phospholipid-Bischicht wird eine stabile und versteifte Membran entstehen, welche zu facettierten Liposomen führen wird.

Wissenschaftlicher und gesellschaftlicher Kontext des Forschungsprojekts 
Unsere Arbeit wird grundlegende neue Erkenntnisse liefern zur Frage, welche Kräfte in natürlichen Zellmembranen wirken. Dadurch wird es möglich, die Kräfte nach Belieben zu steuern und zu verändern. So werden neue Typen von Vesikeln zugänglich, welche z.B. in der Medizin zur Einkapselung von Medikamenten verwendet werden könnten.

Abstract

During the first period of the SNF Professorship, we have made two intriguing observations that we would explore in detail. i) We have developed a new phospholipid synthesis. This versatile synthesis allows the formation of three-tailed phospholipids that should form very tight bilayer membranes. ii) 1,3-Diamido phospholipids for fully interdigitated bilayer membrane leaflets. The hexagonally packed alkyl chains should be exchanged by diacetylene acyl chains. The diacetylenes should then be preorganized for a topochemical polymerization leading to 2D phospholipid bilayer nanosheets.
