The proposed work represents an extension of the previous project „Mitochondrial biogenesis in T. brucei - import of macromolecules“ (31003A_138355). The main emphasis of the proposal will be on mitochondrial protein and mitochondrial tRNA import.-We intend to characterize the protein complex that forms the mitochondrial outer membrane protein translocase in T. brucei and to study the function of its components.-We will extend our studies to the inner membrane and characterize the protein translocase(s) in this compartment. -We will investigate the involvement of the outer and inner membrane protein translocases in mitochondrial tRNA import. To achieve the aims described above we will use various in vivo and in vitro approaches. Moreover we will make use of novel SILAC-based proteomic analyses and determine the mitochondrial proteome across the life cycle of the parasite.
