Lay summary
The aim of this proposal to study the genetic and social mechanisms underlying a fundamental social polymorphism in the fire ant Solenopsis invicta. In the monogyne social form of this species, colonies invariably contain a single queen, whereas colonies of the polygyne form contain anywhere between 2 and 200 queens. This fundamental difference in social organisation is completely associated with variation at the odourant binding protein gene Gp-9. The behaviour of fire ant workers depends on both their own Gp-9 genotype and the proportion of workers harbouring an identical genotype in their colony. We propose to investigate how social interactions together with genomic differences at this locus influence the expression of other genes and, ultimately, affect the behaviour of workers. We will use modern ultrahigh through-put sequencing (UHTS) technologies to sequence the fire ant genome. To obtain information on the genomic region including Gp-9 we will use two haploid males with the different Gp-9 haplotypes. By conducting similar analyses to those performed on sex chromosomes we will gain information on Gp-9 haplotype-specific sequence features such as inversions, deletions, and insertions. In addition, because the same association between Gp-9 genotypic composition of workers and social organisation occurs in the four closest relatives of S. invicta (S. macdonaghi, S. megergates, S. quinquecuspis, and S. richteri -Krieger and Ross 2002) it will be possible to perform comparative analyses to determine the general order in which inversions/recruitment/deletions occurred. These studies should provide information on the different steps that lead to a shift from monogyny to polygyny and thus document for the first time the origin of a genetic social polymorphism.
