Lay summary
The development of metal-based drugs as an alternative strategy in modern drug research is a fast growing discipline. Since rapidly proliferating cells demand high amounts of vitamin B12, the development of inhibitors of B12 dependant enzymes, so called anti-vitamins seems to be very attractive and has not been explored sufficiently enough. The project aims towards the development of a new class of these compounds that develop cytostatic or cytotoxic activity in vivo.Goals to be achieved:1)Synthesis of a new class of modified B12 derivatives2)Optimization of the structure through molecular modelling and chemical synthesis3)Reactivity studies in model reactions and B12 dependant enzymatic reactionsIn conclusion, the main objective of this research proposal is the development of a new class of inhibitors based on an essential and natural molecule.
